Standard course

There is no validated human FOXO4-DRI dose. Preclinical senolytic work used multi-mg/kg intermittent SC schedules in animals. Longevity-community educational pulses often discuss roughly multi-milligram subcutaneous amounts for a few consecutive days, then weeks/months off. Low-mcg daily tables circulating online are alternate recon math only and may not reflect preclinical exposure scale.

Reconstitution steps

  1. Allow the lyophilized 2 mg FOXO4-DRI vial and bacteriostatic water to reach room temperature.
  2. Draw 1.0 mL bacteriostatic water (default for this guide).
  3. Inject BAC slowly down the inner wall of the vial — avoid blasting the powder cake.
  4. Gently swirl or roll until the solution is clear. Do not shake hard (foaming).
  5. Label with recon date and concentration (e.g. "2.0 mg/mL — 1 mL BAC").
  6. Refrigerate at 2–8 °C (35–46 °F), protected from light. Prefer using a reconstituted multi-dose vial within ~28 days with sterile technique.

Research context & considerations

FOXO4-DRI (D-retro-inverso FOXO4 peptide) was designed to selectively clear senescent cells by disrupting FOXO4–p53 survival signaling. Preclinical work (e.g. Baar et al. lineage) is the primary evidence base; research powders are not approved senolytic drugs.
