Overview
P021 (also P21) is a peptidergic compound corresponding to CNTF residues 148–151 with C-terminal adamantylated glycine for BBB permeability and protease resistance. Developed for disease-modifying Alzheimer research, it has extensive 3xTg-AD mouse data showing cognitive rescue, synaptic marker recovery, and pathology reduction — including prenatal-to-postnatal prevention paradigms. No human trials published to date.
Mechanism
Competitively inhibits LIF signaling and increases BDNF transcription/expression; activates TrkB-linked PLC/PKC, MEK/ERK, and PI3K/Akt cascades; reduces tau hyperphosphorylation via GSK-3β pathway effects. Promotes hippocampal neurogenesis and synaptic plasticity.
